SULFUR MAKES NUCLEOTIDES GO AROUND

In my talk I will demonstrate and discuss how replacement of phosphate oxygen atom(s) in nucleotides by sulfur atom(s) to form nucleoside 5’-phosphorothioate, 2’,3’-cyclic phosphorothioate, 5’-phosphorodithioate, or 5’-phosphoroselenothioate, gives rise to unique chemical features and confer beneficial pharmacological properties. I will describe unique chemo-, stereo-, and diastereo- selective syntheses of these novel chemical entities, discuss their mechanism of formation, their distinctive chemical properties, and give examples of their applications as therapeutic agents – e.g. for delaying the progression of Alzheimer’s disease, and for inhibiting the deleterious deposition of calcium pyrophosphate in osteoarthritis/CPPD disease. In addition, I will mention use of these innovative nucleotides as tools for biochemical studies.
